Jeho otec zřeml, když mu bylo jedenáct let. V roce 1853-1856 pracoval jako tiskař v New Yorku, Philadelphii, St. Louis a Cincinnati. V letech 1857-1861 pracoval jako kormidelník na řece Mississippi, což ho inspirovalo k napsání románu Život na Mississippi. Proslavil se zejména tituly Dobrodružství Toma Sawyera a Dobrodružství Huckleberyho Finna.
